If you want to have everything in mono ... sure. If you're just "trying out" the mixes, can I ask why you even need to have a separate headphone output? If you're the only one hearing it, why not just "mix"?

Is it possible to use a DJ splitter cable with Rekordbox DJ (on a Mac). I just want to try out quick mixes on my laptop and not have to connect any controller hardware.

You can use it without hardware connected and just use Key control or a mouse, are you trying to monitor the sound output and send to a mixer?
I'm just using the mouse and keyboard for control. I'm sending the main output to my computer speakers and want an easy way to listen/cue the next track in my headphones.

Got my hands on a Griffin cable and it works with Rekordbox DJ. Settings I used:
Master Output
L: Output 1
R: Output 1
Headphones Output
L: Output 2
R: Output 2
Also tried it WeDJ and that works too with 'Split Output' enabled.
Settings, Audio, Split Output.
