Thats not possible. You will always have to load the Hot Cues and play them ( to get rid of that first latency ). The cue points/loops in memory will always stop.

CDJ 2000 auto hot cue/cue point instant play
I know the question has been asked a billion times about hot cues being auto loaded but what about the cue points. Cue points are automatically loaded and we can call them up when ever we like but what I am asking/wondering is if it is possible to have the cue point instantly play when I push the next or previous cue point thus becoming a hot cue of sorts. Most of the time I forget to load the hot cues but I always store it as a cue point as well so it would be extremely nice if cue points would play instantly as a option when selected. Thank you

I'll forward it as a suggestion for sure but I think its unlikely to become a feature.
